I've been advised that I should add the address 2:250/9999 (for UK) and send an application from this node to 2:250/0 via netmail.
Can someone please give me a little advice regarding the acquisition of a FidoNet address?
I have set my system up for fido netmail (via the Agency HUB 3:770/1), so I assume I have the prerequisites to make an application.

I believe it was me who advised that as you are in the UK, you need to apply in Zone 2. Zone 3 covers Aust/NZ and you generally cannot be
listed in an area in which you do not reside.
I'm not sure who advised you to host-route through 3:770/1. The proper procedure is to send Crash (direct) Netmail from your system to 2:250/0. This demonstrates that you have a correctly-configured mailer online
that can exchange Netmail.
In addition, I believe your email is bouncing mine and ZC2's replies.
This was done whilst I was following Paul Hayton's Mystic tutorials on YouTube. Perhaps I misunderstood some of the material, but don't I need at least one hub in order to send Netmail: 3:770/1 is the only HUB I have the credentials for (via the tutorial)?

That doesn't sound good; can you advised as to a cause/remedy?
